Characterization of a multifunctional regulator for genes involved in maltodextrin transport and metabolism in Thermococcus litoralis

Nach der Entdeckung, dass TrmB der spezifische Transkriptionsregulator (Repressor) des Trehalose/Maltose-Transportgenclusters von T. litoralis ist, werden wir uns auf die neu entdeckte Eigenschaft von TrmB konzentrieren, zusätzliche Gencluster zu kontrollieren, insbesondere ein Maltodextrin verwertendes System und andere Gene, deren Produkte am Zuckerstoffwechsel beteiligt sind. MalK, die ATP hydrolysierende Untereinheit des Trehalose/Maltose-ABC-Transporters, zeigt in seiner regulatorischen Domäne die gleiche Struktur wie das entsprechende MalK-Protein aus E. coli. Im Gegensatz zum E. coli-Protein wechselwirkt MalK aus T. litoralis jedoch nicht mit seinem Transkriptionsregulator. Stattdessen beobachteten wir, dass es Einzelstrang-DNA bindet, die vom Transkript des Operons stammt. Wir werden der Möglichkeit nachgehen, dass MalK die Expression des Operons kontrolliert, vielleicht mit Hilfe von TrmB oder zusätzlicher Proteine.After the discovery of TrmB as the specific transcriptional regulator (i.e., repressor) of the trehalose/maltose transport gene cluster of T. litoralis we will focus on the newly discovered property of TrmB to control additional gene clusters, notably a maltodextrin utilizing system and other genes whose products are related to sugar metabolism. MalK, the ATP-hydrolyzing subunit of the trehalose/maltose ABC transporter has the same regulatory domain structure as the equivalent MalK protein from E. coli. However, unlike the E. coli protein, MalK from T. litoralis does not interact with its transcriptional regulator. Instead, we observed that it binds single-stranded DNA derived from the operon transcript. We will follow the possibility that MalK is controlling the expression of the operon, perhaps with the help of TrmB or additional proteins.
