Structural and spectroscopic analysis of multi-centered metallo enzymes and their functional complexes involved in nitrate- and sulfate-respiration

Redoxreaktionen spielen eine fundamentale Rolle im Energiehaushalt des Lebens. Energiekonservierende Symsteme benützen dazu Kaskaden von Redoxenzymen, wobei Fe und Cu als auch Flavin zu den wichtigsten Aktivzentren gehören, die diese bedeutende Prozesse katalysieren. In dem Projekt soll die Röntgenstrukturanalyse kombiniert mit spektroskopischen Methoden (UV/Vis, EPR/ENDOR, Mössbauer, CD/MCD) eingesetzt werden, um zentrale Bausteine der Nitrat-Atmung (NO3- -- N2/NH3) und der Sulfat-Atmung (So42- -- H2S) zu charakterisieren. Mithilfe der räumlichen Struktur und den spektroskopischen Eigenschaften dieser Module wird ein hochaufgelöstes geometrisches und elektronisches Bild der Aktivzentren entwickelt. Hieraus wird auf die Funktion der Katalysezentren innerhalb eines Moduls geschlossen, ferner werden intra- und intermolekulare Wechselwirkungen definiert. Zielmoleküle sind: (i) der Multihäm-Komplex Cytochrome c Nitritreduktase (NrfAH), (ii) die cd 1 Nitritreduktase, (iii) die TPP-FAD-abhängige Cyclohexan-1,2-dion Hydrolase (erste Α-Ketolase), (iv) die Adenosin-5´-phosphosulfat Reduktase, (v) die Sulfitreduktase, und (vi) der Hmc Komplex.Redox reactions processes play a fundamental role in maintaining life. Practically all biological energy-transducion complexes employ redox cascades. Fe und Cu, and also flavin, are among the most important active centers involved in the mediation and catalysis of these processes. In the project we use a combination of X-ray crystallography and spectroscopy (UV/Vis, EPR/ENDOR, Mössbauer, CD/MCD) to characterize a number of main modules involved in (i) nitrate-respiration (NO3- -- N2/NH3) and (ii) sulfate-respiration (So42- -- H2S). Based on the architecture and spectroscopic features of these building blocks a detailed electronic and magnetic picture of the active sites will be derived which will help in understanding the function of the catalytic centers within one module, and which will help in defining intra- and intermolecular couplings. Prime targets will be: (i) the multiheme complex cytochrome c nitrite reductase (NrfAH), (ii) the cd 1 nitrite reductase, (iii) the TPP-FAD-dependent cyclohexane-1,2-dione hydrolase (first Α-ketolase), (iv) the adenosine-5´-phosposulfate reductase, (v) the sulfite reductase, and (vi) the Hmc complex.
